Clone Unstable DNA
EPICENTRE Biotechnologies’ CopyCutter™ EPI400™ E.
coli cells significantly
lower the copy number of many common vectors so that “toxic” genes
or unstable DNA sequences are easier to clone. The copy
number of vectors containing ColE1- and
pMB1-type origins that are present in various derivatives of pUC,
pBR, pET and many other vectors is reduced up to 25-fold, increasing
the likelihood that an insert will be stabley maintained. The highly
toxic regB gene, for example, was successfully cloned into
a pET11 vector only when recombinants were maintained in CopyCutter
cells.
