The FailSafe™ PCR System combines a unique blend of thermostable
enzymes for high fidelity with an extensively tested set of reaction
PreMixes to provide a powerful system designed to solve all of your PCR
problems. This unique system can be used for consistent amplification
of difficult templates, such as sequences with high GC content or secondary
structure, and long sequences (at least 20 kb), as well as multiplex
PCR.
The FailSafe PCR System includes a unique thermostable polymerase blend
containing a 3' to 5' proofreading enzyme, providing fidelity at least
three times higher than Taq DNA polymerase alone. In addition,
the kit contains convenient FailSafe PCR PreMixes that allow easy determination
of the optimal reaction conditions for a given template/primer pair combination.
These PreMixes contain buffer, dNTPs, and various amounts of MgCl2 and
FailSafe PCR Enhancer (with betaine). Using the FailSafe PCR PreMix Selection
Kit, simply add a mastermix of your template, primers, and the FailSafe
PCR Enzyme Mix to the 12 PreMixes and amplify. Then choose the PreMix
that provides the best results and use it to obtain consistent amplification
every time.
The FailSafe PCR PreMix Selection Kit contains the FailSafe PCR Enzyme
Mix and the 12 FailSafe PCR PreMixes. Once the optimal PreMix has been
determined, that PreMix can be purchased separately or in conjunction
with the FailSafe PCR Enzyme Mix.
